A peptide from the mitochondrial genome
MOTS-c is a 16-residue peptide encoded within the 12S rRNA region of mitochondrial DNA. Its very existence rewrote the assumption that mitochondrial DNA encoded only components of the electron transport chain.
Functionally, MOTS-c behaves as a metabolic signalling molecule: it translocates to the nucleus under metabolic stress and modulates AMPK-dependent gene expression.
Why it belongs in metabolic research
In preclinical work, MOTS-c improves insulin sensitivity and shifts substrate utilisation toward fatty acid oxidation. What makes it interesting is not the direction of the effect but the mechanism — direct engagement of the AMPK pathway without receptor-mediated signalling.
That gives researchers a tool for probing AMPK-driven adaptation in isolation from the incretin and adipokine axes.
Stability
MOTS-c is more sensitive to freeze-thaw cycles than the incretin analogues. Aliquot reconstituted material and minimise repeated warming.
